PLM-2008 is a sample-preparation kit containing 2.0 mL tubes pre-filled with bearing balls and Grinding Buffer for standardized mosquito pool homogenization. It is step 1 in the ProAmpRT arbovirus surveillance workflow. Each kit provides 100 preparations.
Up to 50 mosquitoes per pool. Per the Pro-Lab mosquito protocol IFU (PLM-2004 Ver: 20180706): mosquito pools (n ≤ 50) should be collected into 2.0 mL tubes and kept frozen prior to processing to prevent RNA degradation.
A bead mixer or rotor-stator homogenizer (user-supplied). Per the Pro-Lab mosquito protocol IFU: homogenize 35 seconds at 4,000 rpm, twice; centrifuge 5–10 minutes at 12,000 × g at room temperature; transfer 200 µL of clarified supernatant to the downstream RNA extraction step.
The kit is stored at room temperature with one exception: the Grinding Buffer should be stored at 2–8°C. The kit is stable for at least 12 months under correct storage conditions. Do not freeze any kit components.
After homogenization with PLM-2008 and RNA extraction (PLM-2007 / PLM-2004 / PLM-2000), downstream ProAmpRT LAMP detection kits cover West Nile Virus (PLM-1008), Dengue serotypes 1–4 (PLM-1005), Zika, Chikungunya, Eastern Equine Encephalitis, Western Equine Encephalitis, St. Louis Encephalitis, and La Crosse Encephalitis.
LAMP detects viral RNA; RAMP detects viral antigen. An independent study (Burkhalter et al., J Med Entomol. 2018;55(3):717; PMID: 29462341) evaluated both platforms on honey cards and found LAMP detected WNV and Zika positive samples that RAMP returned as negative. ProAmpRT LAMP also covers EEE, WEE, SLE, LAC, and CHIKV — RAMP does not.
