Test Principle: Polyclonal rabbit antibodies bind to specific somatic (O), flagellar (H), or capsular (K/Vi) antigens on the surface of bacterial cells. Antigen-antibody cross-linking causes visible agglutination (clumping) within 1–2 minutes on a slide. Absence of agglutination indicates no matching surface antigens.

Limitations: Antisera confirm phenotypic antigen expression only. Autoagglutinating or rough strains will agglutinate in saline control and results are not interpretable. Cross-reactivity between antigenically related serotypes may occur. Do not use on mixed cultures.
QC Requirements: Positive and negative controls must be run each day of use per CLIA/CAP requirements. Document lot number, expiry date, and control results per laboratory's QC policy.
Storage: 2–8°C; do not freeze. Cold chain maintained during shipping.
