Normal vaginal pH is acidic — approximately 4.5 to 5.5. Amniotic fluid is neutral to slightly alkaline — approximately 7.0 to 7.5. When membranes rupture and amniotic fluid enters the vaginal vault, it elevates the local pH. AmnioTest™ detects this shift immediately upon withdrawal — a color change visible to the naked eye, compared against the included Color Interpretation Card.
The AmnioTest™ swab is impregnated with nitrazine yellow dye. Insert the swab and observe the color: yellow to olive-green indicates normal vaginal pH and membranes likely intact. Dark green to navy blue at pH 6.5–7.5 indicates possible ruptured membranes. Compare to the included Color Interpretation Card for a standardized, reproducible read.

How does AmnioTest™ detect amniotic fluid?
The swab is impregnated with nitrazine yellow dye. Normal vaginal pH is acidic (~4.5–5.5). Amniotic fluid is neutral (~7.0–7.5). When amniotic fluid is present, vaginal pH rises, causing the dye to shift from yellow/olive to dark green or navy blue at pH 6.5–7.5.
How long does it take to get a result?

Does AmnioTest™ require a speculum?
No. AmnioTest™ does not require speculum placement. This reduces patient discomfort and simplifies the procedure, especially in active labor or triage settings.
What does a positive result look like?
A dark green to navy blue color change indicates pH 6.5–7.5, which is consistent with possible amniotic fluid leakage. Compare the swab color to the included Color Interpretation Card (PL.902). What does a negative result look like?
