Column-based extraction (e.g., Qiagen QIAamp) binds RNA to a silica membrane in a spin column and requires centrifugation at each wash step — typically 3–5 centrifuge spins per extraction. Magnetic bead extraction (Pro-Mag™ PLM-2000) binds RNA to silica-coated beads suspended in solution, then uses a magnetic separator rack to hold the beads against the tube wall while wash buffers are pipetted away. No centrifuge spins. No column membrane to clog. The magnetic bead approach also removes inhibitors (heme, protein, humic acid) more completely from complex sample matrices — which is critical for LAMP, where Bst polymerase is highly sensitive to these inhibitors.

The PLM-2001, PLM-2002, and PLM-2003 are optional magnetic separator racks that complete the no-centrifuge extraction workflow. Without a magnetic rack, you would need a centrifuge to pellet the beads at each wash step. The racks hold the magnetic beads against the tube wall using a permanent magnet, allowing you to pipette off wash buffer manually. LAMP (Loop Mediated Isothermal Amplification) requires higher RNA purity than standard PCR. LAMP uses Bst DNA polymerase (from Bacillus stearothermophilus), which lacks the error-correction mechanism of Taq polymerase and is significantly more sensitive to inhibitors. Heme (from blood), humic acids (from soil/stool), and protein carryover that merely reduce PCR efficiency will completely block LAMP amplification. This is why LAMP-specific extraction validation is a technical requirement, not a marketing claim.
